btn.setBackgroundResource(R.drawable.image);
Relativelayout.addView(btn);

    you can use this :

    //view.findViewById(id); here pass the same id which you have set earlier for a particular view, while adding view to layout/view.
    Button btn = (Button) relativeLayout.findViewById(1);
